PCR amplification of DNA Sequence of Bovine Papillomavirus Type 2 in Urinary Bladder of Cattle with Enzootic Hematuria in Espirito Santo, Brazil

Abstract

The goal of this study was to investigate the presence of bovine papillomavirus type 2 (BPV-2) genomic DNA using polymerase chain reaction (PCR) assays, in the urinary bladder of cattle with bovine enzootic hematuria (BEH), in the neoplastic and non-neoplastic lesions in Espírito Santo, Brazil. Thus, 46 bladders with gross BEH lesions were collected from adult cattle at slaughterhouses. The samples were divided into two portions: one for use in histopathological analysis and the other one for PCR. Microscopic analysis showed that 56.52% of the bladders presented neoplastic lesions, of which 50% were benign, 23.08% were malignant and 26.92% presented both types of neoplasms. The remaining 43.48% of the bladders did not show any neoplasms. Presence of BPV-2 was confirmed in 43.48% of the bladders: 65% of these were found in neoplastic lesions and 35% in non-neoplastic lesions. Among the neoplastic lesions, detection of BPV-2 was more frequent in benign lesions (53.84%), followed by malignant lesions (23.08%) and by those presenting both benign and malignant lesions (23.08%). These results confirm the presence of BPV-2 in both neoplastic and non-neoplastic conditions in urinary bladders of cattle with enzootic hematuria from the studied geographic area.

Isolation, Characterization and Pathology of Citrobacter freundii Infection in Native Brazilian Catfish Pseudoplatystoma

Abstract

Bacterial diseases are responsible for important economic losses in aquaculture over the world, and South American catfishes are susceptible to Gram negative bacteria causative of haemorrhagic septicemia. This study registers the biochemical characteristics, antimicrobial resistance and pathological effects of Citrobacter freundii infection in cacharaPseudoplatystoma reticulatum. Moribund juvenile fish with non-specific clinical signs were used for bacteriological and parasitological diagnosis. No parasitic infections were found in examined fish, but two isolate obtained from the kidney and encephalon was characterized by Gram negative bacilli, catalase positive and oxidase negative. Isolates were submitted to biochemical identification by a commercial API 20E. Susceptibility analysis to 15 drugs was performed by the diffusion method in Agar Muller Hinton discs. Experimental assay was also made to confirm the Kock postulate. The isolates showed biochemical profile corresponding to Citrobacter freundii. A multiresistance at 66.7% of the antibiotic molecules tested was observed. In experimental infection haemorrhagic septicaemia, severe enteritis and important lesions in kidney and gills are described.

Morphological and Immunophenotipical Characterization of Murine Mammary Carcinoma 4t1

Abstract

The 4T1 murine mammary carcinoma is an experimental model widely used in assessing and better understanding of tumor biology. It is a highly tumorigenic cell line and invasive, where metastases are observed in various organs. This study aims to describe morphological and immunophenotipical aspects of 4T1 mammary carcinoma in mice Balb/c with the aid of the immunohistochemistry. Tissues were fixed in formalin and processed using the routine paraffin inclusion technique. Histologic sections (4 µm) were stained through Hematoxylin-Eosin techniques for morphologic assessments. For immunohistochemical study, we used a panel of 9 (nine) antibodies: hormone receptors, receptors for cell proliferation, cytokeratins, vimentin, growth factor receptor and markers of blood vessels. Morphologically, the 4T1 murine mammary carcinoma shows malignant epithelial proliferation in solid arrangement, with pleomorphic cells and high mitotic index. In immunohistochemical analysis was determined a positivity for hormone receptors, cytokeratin AE1/AE3, receptors of cell proliferation and markers of blood vessels. There was a negative for vimentin, cytokeratin 5/6, cytokeratin 34βE12 and growth factor receptor.The results show that the characteristics observed in this model are similar to some types of breast cancers found in women like poorly differentiated invasive ductal carcinoma. Thus, the immunophenotypic characterization of mammary carcinoma 4T1 allows a better understanding of the model to the study of new anticancer therapies.

Case Report
Asteroid Bodies Associated with Granulomatous Meningoencephalitis in a Female Dog

Abstract

A case of asteroid bodies associated with granulomatous meningoencephalitis is reported in a 4-year-old female dachshund that showed intermittent tremors and blindness. Necropsy revealed leptomeninges moderately hyperemic and from the parietal cortex extending into the temporal and frontal cortex, thalamus and midbrain flattening of the gyri and a grayish and friable focally extensive area. Microscopically intense and extensive perivascular inflammatory infiltrates of lymphocytes, plasma cells,macrophages and giant cells were observed. In the cytoplasm of the giant cells, neuropil stellate structures ranging from 20 to 80 μm in diameter with a central dense and homogenous zone and with amphophilic and vacuolated cytoplasm consistent with asteroid bodies were present. Special staining and transmission electron microscopy permitted the diagnosis of asteroid bodies associated with granulomatous meningoencephalitis.

Case Report
Primary Cerebral Hemangiosarcoma in an Adult Labrador Retriever Dog

Abstract

A 6-year-old, castrated male, Labrador Retriever dog presented to the referring veterinarian for acute onset of lethargy. Clinical examination revealed hyperesthesia of the face and blindness in the right eye. The animal died in the hospital and was submitted for postmortem examination. At postmortem examination, a dark red mass (2.0 cm x 1.8 cm x 1.6 cm) had infiltrated the right frontal lobe of the cerebrum.  Microscopically, the tumor was composed of small caliber vascular channels lined by neoplastic endothelial cells. Immunohistochemical staining revealed widespread expression of CD31 and the absence of pan-cytokeratin expression in neoplastic cells. The diagnosis of primary cerebral hemangiosarcoma was made.The brain is a rare primary site for hemangiosarcoma in the dog.

Case Report
Synchronous Multicentric Osteosarcoma in the Canine Axial Skeleton: a Report of Two Cases

Abstract

Two cases of axial synchronous multicentric osteosarcoma are described in male dogs 11 and 13 years of age. In both cases, the necropsy revealed multiple tumor nodulesmasses in the thoracic vertebrae and ribs. The microscopic features indicated that the masses were osteoblastic osteosarcoma for all bone sites with immunohistochemical expression of vimentin and osteocalcin and the absence of cytokeratin. In both cases, multicentric osteosarcoma was considered to be present simultaneously in various bone sites of the axial skeleton, to be of the same histological type and to exhibit the same immunohistochemical characteristics in all bone sites, with no macroscopicmicroscopic metastases to the lungsany other soft tissue.
